Absolute and Multiplex Quantification of Proteins

At BioXpedia we offer protein quantification service using immunoassays from Meso Scale Discovery. In drug discovery and biomarker identification, absolute quantification of proteins can be relevant in order to determine the importance of observed differences in protein levels between patient groups. ELISA is a widely used assay for the quantification of cytokines and other proteins in many clinical samples, but ELISA is cumbersome for multiple analytes.

The Meso Scale Discovery (MSD) technology offers absolute quantitation of protein markers in multiplex using immune assays, which are based on electrochemiluminescence, allowing higher sensitivity than are possible using traditional ELISA.

The MSD technology is highly flexible offering ultra-sensitive assays that can be provided as single analyte assays or multiplex assays up to 10-plex in each well saving precious sample material.

More Than 500 Assays Available

Available MSD assays cover a wide range of biological applications, with more than 500 assays provided in panels, custom panels or reagent kits to build assays.

The biomarker panels are focused on measuring proteins associated to for example inflammation, oncology, neurodegeneration, immunology, intracellular signalling, cardiovascular disease and metabolism. Assays are available for different species and compatible with an array of complex sample types e.g. serum, plasma, supernatants, cell lysates, CSF, urine, blood spots, tears, synovial fluid, tissue extracts etc.

MSD assays are based on a proprietary technology combining electrochemiluminescence and multiarray technology for detection of multiple proteins in biological samples.

Multi-spot microplates allow multiplexing of up to 10 assays in one well, making MSD a rapid technology for multiplex protein quantification.

Electrochemiluminescence detection in addition offers a unique combination of sensitivity, accuracy and dynamic range for absolute quantitation of single proteins or protein profiles.

Validated Protein Quantification Assays

V-PLEX assays are designed to maximize reproducible performance, consistency and reliability in results and confidence in data. The assays and panels are fully validated in multiple matrices according to fit-for-purpose principles. The validated assays come in both separate assays or multiplex variants and are designed to ensure optimal and consistent performance and lot-to-lot consistency is guaranteed to support long-term studies.

Custom Single Protein or Multiplex Immunoassays

The U-PLEX assays offer full flexibility to custom design your own singleplex or multiplex assays. MSD has developed U-PLEX assays allowing you to study the desired proteins unique for your project.

In the U-PLEX assays the specific capture antibodies are coupled to U-PLEX linkers, which bind to spots on the plate. Analytes in the sample will bind to the capture antibody, and then detection antibodies conjugated with electrochemiluminescence labels bind to the analytes to complete the sandwich assay.

Finally, R-PLEX antibody sets can also be used for development of multiplex or single analyte immunoassays. Each R-PLEX antibody set contains a biotinylated capture antibody, electrochemiluminescence labelled detection antibody and a recombinant protein for standard.
